①Heparinases

Heparinases can clave glycosidic bonds of heparin and/or heparan sulfate by a β-elimination mechanism, generating unsaturated products (mostly 

disaccharides) with a double bond be-tween C4 and C5 of the uronate residue.

More Information

Alpha-thrombin is a product of calf plasma prothrombin activated by FXa (assisted by FVa and phospholipids), purified by ion exchange and gel filtration chromatography, and inactivated by S/D virus.

Thrombin, also known as Factor IIa (FIIa), plays a very important role in the coagulation waterfall as a serine protease.

The Alpha-thrombin reagent produced by Beijing Asler Biotechnology Co., Ltd. is specially optimized for the detection of heparin anti-FIIa titer, and the main configuration is Alpha-thrombin, which is very beneficial to the detection method due to its high purity and high stability.

How to use

 

Add an appropriate amount of water or buffer to obtain the desired concentration of Alpha-thrombin solution according to application requirements.

 

Stability and storage methods

 

Alpha-thrombin lyophilized products should be stored at 2-8°C in a dark place and used within the expiration date indicated on the label;

5 IU/mL Alpha-thrombin solution is stored at 2-8°C in a dark place for about 7 days.

Alpha-thrombin is suitable for the detection of anti-FIIa activity of anticoagulant drugs such as heparin, and is only used for experimental research and industrial quality control, and cannot be used as a drug or in vivo experiment.

Alpha-thrombin has a wide range of uses, such as

  • The ratio of anti-FIIa activity and anti-FXa activity of unfractionated heparin and low molecular weight heparin was detected
  • Accurate detection of anti-FIIa activity of unfractionated heparin and low molecular weight heparin
  • Monitor the amount of heparin in blood samples

About the color substrate method

 

Alpha-thrombin (FXa) can be used to detect the anti-FIIa activity of heparin (chromogenic substrate method).

Chromogenic substrates are small chemically synthesized peptides with chromogenic groups (pNA) at one end. Specific enzymes (such as thrombin) catalyze to dissociate pNA. Free pNA can be detected by a spectrophotometer or microplate reader to determine the activity of the corresponding substance.

Common chromogenic substrates are: thrombin chromogenic substrate S-2238, FXa chromogenic substrate S-2765 and S-2222.

Chromogenic assays are currently the main methods for detecting heparin titers, which are the methods for the detection of heparin (especially low molecular weight heparin) specified in the European Pharmacopoeia (EP) and the United States Pharmacopoeia (USP). This method is also widely used in laboratory research and in vitro diagnostics.

By controlling the conditions of the reaction system, chromogenic substrate methods can detect clotting factors, cofactors, or clotting factor inhibitors.
